Ever since our neighbours moved in next door there seems to be a smell of smoke in the upstairs room that is closest to the neighbour. It smells like cigarette smoke. If it is from them is it possible to do anything with this vent other than block it which seems like the wrong thing to do?

Yes. There is.
Search for a ‘ducting cowled gravity flap’. Get some adhesive and stick it to this vent (you may need to snap some parts off the vent you buy to make it flush) and use silicon on the sides to seal them up.

My vent is identical to that. I put expandable foam but that wasn’t a good idea.
