AstraZeneca Pharma India Ltd. (AZPIL) announced that the Central Drugs Standard Control Organisation (CDSCO) has approved an\u00a0additional indication for Trastuzumab Deruxtecan 100 mg\/5 mL vial lyophilized powder for concentrate for solution for infusion, in combination with\u00a0Pertuzumab\u00a0as a first-line treatment for adult patients with\u00a0unresectable or metastatic HER2-positive (IHC 3+ or ISH+) breast cancer.<\/p>\n
In India,\u00a0HER2-positive breast cancer, defined by HER2 gene amplification or protein overexpression, is estimated to account for approximately 12% to 19% of breast cancer cases. In the metastatic setting, unmet need remains significant. Despite advances in care, no new first-line therapies have enhanced outcomes for patients with HER2-positive metastatic breast cancer in recent years. In addition, approximately\u00a0one-third of patients\u00a0treated in the first-line setting do not go on to receive second-line therapy, highlighting the importance of using the more effective treatment early in the course of the disease.<\/p>\n
The approval is based on results from the\u00a0DESTINY-Breast09\u00a0trial, which included patients from India.\u00a0In this trial, Trastuzumab Deruxtecan plus Pertuzumab\u00a0demonstrated a statistically significant and clinically meaningful improvement in\u00a0progression-free survival (PFS)\u00a0by\u00a0blinded independent central review (BICR)\u00a0as compared to the THP (taxane, trastuzumab and pertuzumab) regime. Median PFS was\u00a040.7 months\u00a0for Trastuzumab Deruxtecan plus Pertuzumab compared with\u00a026.9 months\u00a0for THP, representing a\u00a044% reduction in the risk of disease progression or death. The benefit was consistently observed across subgroups.<\/p>\n

Current approved indications for trastuzumab deruxtecan\u00a0include the treatment of adult patients with unresectable or metastatic\u00a0HER2-positive breast cancer\u00a0who have received a prior anti-HER2-based regimen;\u00a0HER2-low\u00a0and\u00a0HER2-ultralow breast cancer\u00a0in specified metastatic settings;\u00a0locally advanced or metastatic HER2-positive gastric or gastroesophageal junction adenocarcinoma\u00a0following a prior anti-HER2-based regimen; and\u00a0other unresectable or metastatic HER2-positive (IHC 3+) solid tumours\u00a0after prior systemic treatment where no satisfactory alternative treatment options exist.<\/p>\n
