The damage to AstraZeneca PLC (LSE:AZN, NASDAQ:AZN)<\/a> from Wainua’s trial failure is less about the numbers than about a dent to management’s credibility, according to Jefferies. <\/p>\n

The shares fell 9% and topped the FTSE 100 fallers’ list, a move the broker expects to exceed the drug’s actual contribution to valuation. <\/p>\n

Jefferies, which rates the stock a ‘buy’ with an 18,000p target, forecasts peak Wainua sales in the heart indication of $4 billion at a 60% probability. <\/p>\n

That equates to around 250p a share of risk-adjusted value, or roughly 2% of net present value. <\/p>\n

The bank’s blunt assessment is that the initial share price move will likely be double that impact. <\/p>\n

The reason lies in expectations. AstraZeneca is regarded as capable of designing near-watertight trials, so a failure raises awkward questions. <\/p>\n

Jefferies pointed to a specific culprit: a 24% drop-in of rival drug tafamidis during the study, which lifted the share of patients on the competing treatment from 57% at the outset towards around 80%. <\/p>\n

That is well above the 53% seen in Pfizer’s rival HELIOS-B trial, muddying Wainua’s chance of showing benefit on top of existing therapy. <\/p>\n

Encouragingly, patients on Wainua alone showed a 29% reduction in events, a nominally significant result broadly in line with HELIOS-B. <\/p>\n

Management had been highly confident of hitting the primary endpoint and demonstrating benefit in combination use, making the miss harder to explain away. <\/p>\n

The timing compounds the problem, the broker suggested. <\/p>\n

Investors have little appetite for volatility events, and the next major catalyst, the AVANZAR lung cancer readout expected around July or August, is itself contested. <\/p>\n

Jefferies suspects the stock may not recover until that overhang clears. <\/p>\n
