Chinese innovative drugmaker Simcere Pharmaceutical (2096.HK) has entered into a global exclusive licensing collaboration with Swiss pharmaceutical giant Roche, centered on the trispecific antibody SIM0660, with a potential total transaction value of up to $1.53 billion. Following the announcement, Simcere Pharmaceutical’s Hong Kong-listed shares opened nearly 5% higher on September 2, trading at HK$11.52, with gains reaching 4.82% at one point and turnover of approximately HK$1.8893 million.

The agreement was signed between Simcere Zaiming, Simcere Pharmaceutical’s oncology-focused subsidiary, and Roche. Under the terms, Roche will obtain exclusive worldwide rights to develop, manufacture, and commercialize SIM0660. Simcere Zaiming is eligible to receive cumulative payments of up to $1.53 billion, including a $75 million upfront payment. In addition, the company will be entitled to tiered royalties of up to double-digit percentages on future net product sales.

Dual-Targeting Design Aimed at B-Cell Diseases

SIM0660 is a trispecific antibody targeting CD79a, CD19, and CD3, developed by Simcere Zaiming using its proprietary T-cell engager (TCE) multispecific antibody technology platform. The molecule’s design features a CD3-binding T-cell engager arm fused with two binding domains that respectively target the B-cell surface antigens CD79a and CD19, thereby inducing potent T-cell-mediated cytotoxicity while effectively suppressing cytokine release.

This dual-targeting strategy that simultaneously engages CD79a and CD19 holds potential first-in-class status among existing therapies. SIM0660 is designed to provide broader therapeutic coverage for various B-cell-mediated diseases, potentially offering differentiated treatment options for patients who have responded poorly to prior CD20 or CD19-targeted therapies. Beyond oncology, the molecule is also expected to show promise in B-cell-mediated autoimmune diseases, with the potential to deliver clinical benefits through the specific depletion of pathogenic B cells.

Cumulative Out-Licensing Value Surpasses $6.1 Billion

This transaction represents the latest achievement in Simcere Pharmaceutical Group’s ongoing international licensing strategy. To date, the group has completed six out-licensing deals, with cumulative potential total consideration exceeding $6.1 billion, demonstrating that its R&D capabilities in innovative antibody therapeutics have gained increasing recognition from global pharmaceutical leaders.

From a market reaction standpoint, investors have responded positively to the collaboration. Simcere Pharmaceutical’s share price strengthened significantly on the first trading day following the announcement, reflecting market optimism about the company’s pipeline value and future licensing revenue potential. For Roche, bringing in SIM0660 further strengthens its product portfolio in B-cell-related diseases, particularly in seeking greater strategic flexibility across the two major therapeutic areas of hematological malignancies and autoimmune diseases.
