{"id":81403,"date":"2026-06-11T10:13:23","date_gmt":"2026-06-11T10:13:23","guid":{"rendered":"https:\/\/www.europesays.com\/ch\/81403\/"},"modified":"2026-06-11T10:13:23","modified_gmt":"2026-06-11T10:13:23","slug":"novartis-jettisons-ph-1-cancer-candidate-after-0-efficacy","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/ch\/81403\/","title":{"rendered":"Novartis jettisons ph. 1 cancer candidate after 0% efficacy"},"content":{"rendered":"<p>Novartis has jettisoned an anti-cancer protein from its pipeline after the molecule failed to shrink tumors in 30 patients with advanced disease.<\/p>\n<p>The pharma giant halted the <a href=\"https:\/\/clinicaltrials.gov\/study\/NCT05462873\" rel=\"nofollow noopener\" target=\"_blank\">phase 1 trial<\/a> of QEQ278 and has dropped the candidate entirely, a spokesperson confirmed with Fierce Biotech.\u00a0<\/p>\n<p>Novartis had <a href=\"https:\/\/aacrjournals.org\/mct\/article\/24\/10_Supplement\/B002\/766263\" rel=\"nofollow noopener\" target=\"_blank\">previously shared<\/a> early data from the trial showing that none of the patients treated with the protein achieved an objective response, with 26 of them halting treatment partway through the trial because their cancer got worse. Patients included in the trial had advanced malignancies and exhausted standard of care treatments.<\/p>\n<p>\u201cFollowing review in 2024, Novartis made the decision to discontinue development of QEQ278 in the phase 1 study,\u201d the spokesperson told Fierce. \u201cThis decision was not based on any safety concern and the asset was removed from the Novartis pipeline.\u201d<\/p>\n<p>Despite the discontinuation occurring years ago, the trial\u2019s status was only just updated \u201cper protocol\u201d after \u201cthe remaining participant recently discontinued study treatment,\u201d the spokesperson added.<\/p>\n<p>News of the terminated trial was first reported by ApexOnco.\u00a0<\/p>\n<p>QEQ278 was designed to target a group of proteins that bind to the natural killer group 2D receptor (NKG2D) on the surface of natural killer (NK) cells. Many tumor cells are <a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC6145810\/\" rel=\"nofollow noopener\" target=\"_blank\">littered with ligands<\/a> that bind to NKG2D, which normally, the natural killer cells, could use to identify the cancer and destroy it. As a decoy, these cancers tend to shed some of the ligands into the bloodstream, which distracts the immune cells and allows the tumor to evade detection.<\/p>\n<p>With QEQ278, Novartis hoped to both nullify the decoy ligands floating in the body and also attach to the surface of tumor cells and flag them for destruction.<\/p>\n<p>Other companies have also tried to target NKG2D ligands, with limited success. For example, Nkarta engineered NK cells to express a boosted NKG2D receptor and saw some early wins in blood cancer, but ultimately <a href=\"https:\/\/www.fiercebiotech.com\/biotech\/nkartas-blood-cancer-car-stalls-again-falling-response-rate-forcing-it-out-race\" rel=\"nofollow noopener\" target=\"_blank\">dropped the candidate<\/a> in March 2024 after the response rate plummeted when more patients were treated.<\/p>\n<p>Astellas <a href=\"https:\/\/www.fiercebiotech.com\/biotech\/astellas-calls-it-quits-early-stage-lymphoma-car-t-following-pipeline-review\" rel=\"nofollow noopener\" target=\"_blank\">similarly canned<\/a> an early CAR-T lymphoma candidate using NKG2D in January 2025, which a company spokesperson told Fierce at the time was a \u201cdifficult decision\u201d driven by \u201cprioritization efforts.\u201d<\/p>\n<p>Novartis\u2019 decision coincides with Biogen discontinuing a combination arm of a <a href=\"https:\/\/clinicaltrials.gov\/study\/NCT05798520\" rel=\"nofollow noopener\" target=\"_blank\">phase 2 trial<\/a> for multiple sclerosis candidate BIIB091.
