A tentokr\u00e1t je v hlavn\u00ed roli \u010cech. \u00a0<\/p>\n<p>Zat\u00edm jen ve form\u011b preprintu na platform\u011b bioRxiv zve\u0159ejnil t\u00fdm veden\u00fd Dieterem Eglim z\u00a0newyorsk\u00e9 Columbia University v\u00fdsledky experiment\u016f, p\u0159i nich\u017e v\u011bdci c\u00edlen\u011b upravili v\u00a0lidsk\u00fdch embry\u00edch trojici gen\u016f. Je kolem toho celkem poprask. Pro n\u00e1s m\u016f\u017ee cel\u00e1 kauza z\u00edskat na zaj\u00edmavosti i d\u00edky prvn\u00edmu autorovi studie \u0160t\u011bp\u00e1novi Je\u0159\u00e1bkovi, kter\u00fd se krom\u011b p\u016fsoben\u00ed na Columbia University hl\u00e1s\u00ed jako k\u00a0domovsk\u00e9 organizaci k\u00a0\u00dastavu organick\u00e9 chemie a biochemie AV \u010cR. Dal\u0161\u00ed \u010deskou stopu reprezentuje Marcos Iuri Roos Kulmann, kter\u00fd p\u016fsob\u00ed na \u00dastavu molekul\u00e1rn\u00ed genetiky AV \u010cR a na Karlov\u011b univerzit\u011b.<\/p>\n<p>\u00a0<\/p>\n<p>O co jde? Rozhodn\u011b ne o skand\u00e1l \u010d\u00ednsk\u00e9ho kalibru z\u00a0roku 2018, kdy \u010d\u00ednsk\u00fd biofyzik Jiankui He zas\u00e1hl do d\u011bdi\u010dn\u00e9 informace t\u0159\u00ed lidsk\u00fdch embry\u00ed, z\u00a0kter\u00fdch se narodily d\u011bti. \u010casopis Time za\u0159adil n\u00e1sleduj\u00edc\u00ed rok \u010c\u00ed\u0148ana mezi 100 nejvlivn\u011bj\u0161\u00edch lid\u00ed planety, ale odborn\u00fd sv\u011bt byl z\u00a0valn\u00e9 \u010d\u00e1sti zhnusen a \u010d\u00ednsk\u00e1 justice mu vym\u011b\u0159ila za poru\u0161en\u00ed z\u00e1kon\u016f 3 a p\u016fl roku natvrdo. Jiankui He pou\u017eil pro z\u00e1sah do d\u011bdi\u010dn\u00e9 informace technologii CRISPR-Cas9 a ta je pro tyto \u00fa\u010dely m\u00e1lo spolehliv\u00e1. M\u016f\u017ee v\u00a0d\u011bdi\u010dn\u00e9 informaci embrya nap\u00e1chat v\u00edc \u0161kody ne\u017e u\u017eitku. Jasn\u011b se to uk\u00e1zalo nap\u0159. v\u00a0experimentech, kdy se v\u011bdci z\u00a0t\u00fdmu \u0160ukrata Mitalipova pokusili v\u00a0oplozen\u00e9m vaj\u00ed\u010dku korigovat vadn\u00fd gen a v\u00fdsledkem byly ztr\u00e1ty rozs\u00e1hl\u00fdch oblast\u00ed genomu v\u00a0takto vznikl\u00fdch embry\u00edch. To byly d\u016fvody, pro\u010d \u0160t\u011bp\u00e1n Je\u0159\u00e1bek p\u0159istoupil k\u00a0z\u00e1sah\u016fm do d\u011bdi\u010dn\u00e9 informace lidsk\u00fdch embry\u00ed \u00fapln\u011b jinak. Nic z\u00a0toho ale nen\u00ed bez rizika. Ke st\u0159ih\u016fm toti\u017e m\u016f\u017ee doch\u00e1zet i mimo c\u00edlov\u00e9 m\u00edsto, co\u017e hroz\u00ed vznikem genetick\u00e9ho defektu. Zd\u00e1 se, \u017ee d\u011bdi\u010dn\u00e1 informace lidsk\u00e9ho embrya je k\u00a0takov\u00fdm \u201enec\u00edlov\u00fdm st\u0159ih\u016fm\u201c zvl\u00e1\u0161t\u011b n\u00e1chyln\u00e1.<\/p>\n<p>\u00a0<\/p>\n<p>P\u0159i base editingu nen\u00ed dvojit\u00e1 \u0161roubovice st\u0159\u00edh\u00e1na. Na vybran\u00e9 m\u00edsto d\u011bdi\u010dn\u00e9 informace je dopraven enzym, kter\u00fd katalyzuje biochemickou reakci, a tou se m\u011bn\u00ed jedno p\u00edsmeno genetick\u00e9ho k\u00f3du na jin\u00e9 p\u00edsmeno. Je\u0159\u00e1bek pou\u017eil enzym, kter\u00fd m\u011bn\u00ed adenin na guanin. Base editingem pak navodil zm\u011bny ve t\u0159ech genech lidsk\u00fdch embry\u00ed. V\u00a0genu PCSK9 vytvo\u0159il \u201ete\u010dku\u201c, kter\u00e1 nedovol\u00ed, aby se podle genu vyrobila p\u0159\u00edslu\u0161n\u00e1 b\u00edlkovina. Gen PCSK9 hraje d\u016fle\u017eitou roli v\u00a0metabolismu cholesterolu. Jeho vy\u0159azen\u00edm z\u00a0\u010dinnosti lze sn\u00ed\u017eit hladiny cholesterolu v\u00a0krvi u lid\u00ed, kter\u00e9 ohro\u017euj\u00ed extr\u00e9mn\u011b vysok\u00e9 hodnoty cholesterolu a nezab\u00edraj\u00ed jim l\u00e9ky.<\/p>\n<p>D\u00e1le se Je\u0159\u00e1bek a spol. zam\u011b\u0159ili na geny HBG1 a HBG2. Ty k\u00f3duj\u00ed tzv. fet\u00e1ln\u00ed hemoglobin obsa\u017een\u00fd v \u010derven\u00fdch krvink\u00e1ch lidsk\u00e9ho embrya a plodu. Po narozen\u00ed jsou tyto geny \u201eusp\u00e1ny\u201c a fet\u00e1ln\u00ed hemoglobin je v\u00a0\u010derven\u00fdch krvink\u00e1ch nahrazen \u201edosp\u011bl\u00fdm\u201c hemoglobinem A. Pokud m\u00e1 \u010dlov\u011bk po\u0161kozen n\u011bkter\u00fd z\u00a0gen\u016f pro tvorbu dosp\u011bl\u00e9ho hemoglobinu A, m\u016f\u017ee trp\u011bt d\u011bdi\u010dn\u00fdmi chorobami, jako je srpkovit\u00e1 an\u00e9mie, beta-talasemie aj.<\/p>\n<p>\u00a0<\/p>\n<p>Dnes u\u017e se vyv\u00edjej\u00ed v\u00a0r\u00e1mci tzv. genov\u00fdch terapi\u00ed z\u00e1sahy do d\u011bdi\u010dn\u00e9 informace t\u011bln\u00edch bun\u011bk pacient\u016f pomoc\u00ed CRISPR, kter\u00e9 c\u00edl\u00ed na l\u00e9\u010dbu vysok\u00fdch hladin cholesterolu blokov\u00e1n\u00edm genu PCSK9. Obnova tvorby fet\u00e1ln\u00edho hemoglobinu genovou terapi\u00ed Casgevy u\u017e byla schv\u00e1lena jak pro l\u00e9\u010dbu srpkovit\u00e9 anemie, tak i beta-talasemie. \u0160t\u011bp\u00e1n Je\u0159\u00e1bek se rozhodl uk\u00e1zat, jak by podobn\u00e9 z\u00e1sahy s\u00a0vyu\u017eit\u00edm base editingu zab\u00edraly v\u00a0lidsk\u00e9m embryu. V\u00fdsledky jsou bezesporu zaj\u00edmav\u00e9. Z\u00e1rove\u0148 ale nejsou nijak povzbudiv\u00e9. To znamen\u00e1, \u017ee z\u00e1sah do DNA pomoc\u00ed base editingu se neprojevil ve v\u0161ech bu\u0148k\u00e1ch embrya. N\u011bkde z\u016fstala p\u016fvodn\u00ed konfigurace DNA a p\u00edsmeno genetick\u00e9ho k\u00f3du adenin se nep\u0159epsalo na guanin. To je nep\u0159\u00edjemn\u00e9, proto\u017ee v\u00a0dal\u0161\u00edm v\u00fdvoji se mohou bu\u0148ky s\u00a0nezm\u011bn\u011bnou d\u011bdi\u010dnou informac\u00ed pod\u00edlet na stavb\u011b tk\u00e1n\u00ed a org\u00e1n\u016f. Choroba, jako je vysok\u00fd cholesterol, srpkovit\u00e1 anemie nebo beta-talasemie, se pak m\u016f\u017ee r\u016fznou m\u011brou p\u0159eci jen projevit.<\/p>\n<p>Tak\u00e9 se uk\u00e1zalo, \u017ee z\u00e1sah base editingem je pom\u011brn\u011b komplikovan\u00fd. V\u011bdc\u016fm se nab\u00edz\u00ed, aby ho do embrya vnesli v\u00a0podob\u011b kyseliny ribonukleov\u00e9 (RNA) a ponechali embryo, aby si pot\u0159ebn\u00e9 b\u00edlkovinn\u00e9 n\u00e1stroje pro tyto \u00fa\u010dely samo vytvo\u0159ilo podle dodan\u00e9 RNA. Jen\u017ee v\u00a0tomto p\u0159\u00edpad\u011b reaguj\u00ed embrya na d\u00e1vku RNA zastaven\u00edm v\u00fdvoje. L\u00e9pe se jim vedlo, kdy\u017e do nich v\u011bdci dodali n\u00e1stroj pro base editing hotov\u00fd ve form\u011b proteinu. V\u00fdroba takov\u00e9ho n\u00e1stroje je ale mnohem komplikovan\u011bj\u0161\u00ed a tak\u00e9 dra\u017e\u0161\u00ed.<\/p>\n<p>D\u016fle\u017eit\u00e9 je i zji\u0161t\u011bn\u00ed, \u017ee i base editing m\u011bl \u201enec\u00edlov\u00e9 z\u00e1sahy\u201c. Tam, kde m\u011bl CRISPR tendenci k\u00a0nec\u00edlov\u00fdm st\u0159ih\u016fm, tam podobn\u011b konstruovan\u00fd n\u00e1stroj pro base editing p\u0159episoval \u201enav\u00edc\u201c adeniny na guaniny.
