K\u00f5rvaltoimete risk on suurem neil, kes kasutavad samaaegselt mitut ravimit.<\/p>\n<p>Fookuses kaks olulist maksaens\u00fc\u00fcmi<\/p>\n<p>Ravimid ei m\u00f5ju k\u00f5igile \u00fchtemoodi ning Tartu \u00dclikooli teadlaste ning Kliinikumi ja P\u00f5hja-Eesti Regionaalhaigla arstide koost\u00f6\u00f6s valminud teadusartikkel n\u00e4itab, et p\u00f5hjuseks v\u00f5ib olla ravimite ja geenide koosm\u00f5ju.<\/p>\n<p>Uuring keskendus kahele olulisele maksaens\u00fc\u00fcmile \u2013 CYP2C19 ja CYP2D6 \u2013, mis osalevad ligikaudu 40 protsendi kliinilises kasutuses olevate ravimite lagundamises. Teadlased tuvastasid CYP2C19 geenis kaks varem uurimata, erineva pikkusega puuduvat DNA l\u00f5iku. Selgus, et nende geenivariantidega inimestel v\u00f5ib ravimite ainevahetus olla oluliselt aeglasem kui tavap\u00e4raselt.<\/p>\n<p>Farmakogenoomika teaduri Kristi Krebsi s\u00f5nul hakkab tavaliselt ravimi sisaldus organismis p\u00e4rast imendumist j\u00e4rk-j\u00e4rgult v\u00e4henema, kuid uuritud geenimuutustega inimeste puhul see sageli nii ei ole. \u00abLeidsime, et neil inimestel ei toimi maksaens\u00fc\u00fcm ning ravim ei lagune ootusp\u00e4rasel kiirusel, vaid v\u00f5ib organismi kuhjuda. See t\u00e4hendab olenevalt ravimit\u00fc\u00fcbist ravimi toime halvenemist v\u00f5i suuremat k\u00f5rvaltoimete riski isegi tavap\u00e4raste ravimiannuste juures,\u00bb selgitas Krebs.<\/p>\n<p>Geneetilisest testist \u00fcksi ei piisa<\/p>\n<p>Uuring t\u00f5i esile ka olulise t\u00f5siasja: ainu\u00fcksi geneetiline test ei pruugi alati peegeldada ens\u00fc\u00fcmi tegelikku aktiivsust inimese kehas. Teadlased n\u00e4itasid, et ravimite lagundamist v\u00f5ivad m\u00e4rkimisv\u00e4\u00e4rselt m\u00f5jutada ka mittegeneetilised tegurid.<\/p>\n<p>\u00abMeie uuring selgitab, et ens\u00fc\u00fcmide t\u00f6\u00f6d v\u00f5ivad oluliselt m\u00f5jutada ka erinevate ravimite samaaegne kasutamine, aga rolli m\u00e4ngivad ka haigused ja elustiil,\u00bb \u00fctles Krebs. Uuringust selgus, et ens\u00fc\u00fcme p\u00e4rssivate ravimite kooskasutamine aeglustas ravimite lagundamist ka neil inimestel, kelle geneetiline profiil viitab muidu tavap\u00e4rasele ainevahetusele.
