Ciampa<\/a> sees pregnant patients experiencing preterm labor every day. \u201cIt\u2019s always struck me that for these patients, we have nothing to offer them,\u201d she says. There are no existing therapies that will change the outcome of their pregnancy in the majority of cases, she says, \u201cbut also no good explanation for most cases why the patient is experiencing preterm labor.\u201d<\/p>\n<p class=\"article-content\">Now researchers led by Ciampa and <a href=\"https:\/\/labs.utsouthwestern.edu\/parikh-lab\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">Samir M. Parikh<\/a> at the University of Texas Southwestern Medical Center have linked depletion of nicotinamide adenine dinucleotide (NAD+) in <a href=\"https:\/\/cen.acs.org\/articles\/92\/i32\/Human-Placenta-Project.html\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">the placenta<\/a> to the onset of labor. In studies in mice and in human placentas from healthy pregnancies, researchers have found that <a href=\"https:\/\/doi.org\/10.1126\/science.adz1624\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">labor begins as placental NAD+ levels decrease<\/a> (Science 2026, DOI: 10.1126\/science.adz1624).<\/p>\n<p class=\"article-content\">Parikh says there\u2019s a huge potential impact from investigating the causes of preterm labor. \u201cNormal gestational length is so critical for the long-term health of the baby.\u201d<\/p>\n<p>              <img data-lazy-src=\"https:\/\/www.europesays.com\/ie\/wp-content\/uploads\/2026\/06\/News---Embargo-June-11-2-pm---NAD--in-the-placenta-coordinates-labor---500247.webp\"  alt=\"Structure of NAD&lt;sup&gt;+&lt;\/sup&gt;.\" class=\"w-100\" decoding=\"async\"\/><br \/>\n              Structure of NAD+.<\/p>\n<p class=\"article-content\">The team zeroed in on NAD+ after doing a global metabolomic study looking at placentas from healthy mouse pregnancies. They found that a decrease in levels of the coenzyme coordinated the onset of labor. What\u2019s more, supplements containing NAD+ precursors were able to rescue premature births in mice.<\/p>\n<p class=\"article-content\">NAD+ is vital to many biochemical processes in the body. One role is as a cofactor for the enzyme 15-hydroxy prostaglandin dehydrogenase, which oxidizes certain prostaglandins to their biologically inactive keto forms. \u201cWhen NAD+ is abundant, it\u2019s acting as a brake on labor by helping this enzyme inactivate those prostaglandins, which would otherwise promote labor onset,\u201d Ciampa explains.<\/p>\n<p class=\"article-content\">Ciampa says the ultimate goal of the research is to see if the pathway can be used therapeutically to halt preterm labor or diagnostically to predict which patients are at risk for preterm labor. But, she says, there are many questions left to answer. \u201cAre NAD+-boosting supplements safe? Are they effective at modulating NAD+ levels in the placenta? And does that have positive impacts on a pregnancy?\u201d she asks.
