{"id":591226,"date":"2026-07-17T23:38:31","date_gmt":"2026-07-17T23:38:31","guid":{"rendered":"https:\/\/www.europesays.com\/ie\/591226\/"},"modified":"2026-07-17T23:38:31","modified_gmt":"2026-07-17T23:38:31","slug":"dichloromethane-can-act-as-an-amide-coupling-reagent","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/ie\/591226\/","title":{"rendered":"Dichloromethane can act as an amide coupling reagent"},"content":{"rendered":"<p class=\"article-content\">\u00a0<\/p>\n<p class=\"article-content\">In a chemistry version of whipping up a delicious dinner from pantry staples, <a href=\"https:\/\/doi.org\/10.1021\/jacs.6c05971\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">researchers in Korea have found a recipe for forging amide bonds<\/a> using inexpensive chemicals stocked in nearly any organic laboratory: dichloromethane (DCM)\u2014also called methylene chloride and best known as a workhorse polar aprotic solvent\u2014and a basic salt (J. Am. Chem. Soc. 2026, DOI: 10.1021\/jacs.6c05971).<\/p>\n<p class=\"article-content\">\u201cIt\u2019s something that most people could walk in their lab and do without even bothering to buy anything,\u201d says <a href=\"http:\/\/www.tomsheppard.info\/\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">Tom Sheppard<\/a>, an organic chemist at University College London who was not involved in the work.<\/p>\n<p class=\"article-content\">Amide linkages are important structural motifs in organic chemistry; they appear in the backbones of proteins, polymers, small-molecule drugs, and more. <a href=\"https:\/\/cen.acs.org\/synthesis\/biocatalysis\/Engineered-enzyme-makes-amides-aldehydes\/104\/web\/2026\/02\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">New ways to make them<\/a> more inexpensively and efficiently have a lot of potential for impact.<\/p>\n<p class=\"article-content\">Chonnam National University\u2019s <a href=\"https:\/\/sites.google.com\/view\/sunlabsince2004\/members\/pi\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">Sunwoo Lee<\/a>, who led the work, says his graduate student Nithin Pootheri discovered the reactivity while working on an iridium-catalyzed decarboxylation reaction with DCM as the solvent. The intended reaction failed but produced chlorinated molecules that could undergo substitution with an amine to make an amide, the researchers found. The amide formation happened even without the metal catalyst, thus indicating that DCM was responsible for the reaction.<\/p>\n<p class=\"article-content\">\u201cAt the beginning, I didn\u2019t believe the result,\u201d Lee says. It seemed almost unthinkable that nobody had noticed this behavior from DCM before, but further investigations into the mechanism convinced him.<\/p>\n<p class=\"article-content\">The researchers found that the reaction works on a moderate variety of carboxylic acid and alkyl amine starting materials. It usually preserves the starting materials\u2019 stereochemistry, though amino acids showed a slight tendency to scramble under the basic conditions. The reaction also struggles with sterically bulky amines and doesn\u2019t work at all with aromatic amines, which Lee says he and his team are working to remedy.<\/p>\n<p class=\"article-content\">DCM has a number of health hazards, including cancer from long-term exposure, that have led to <a href=\"https:\/\/cen.acs.org\/policy\/chemical-regulation\/EPA-bans-uses-methylene-chloride\/102\/web\/2024\/04\" shape=\"rect\" rel=\"nofollow noopener\" target=\"_blank\">the solvent being tightly regulated<\/a> in the US and European Union. Many companies are phasing it out of their processes. Lee acknowledges this as a potential obstacle for future industrial applications, but he says that the DCM method could still have process advantages over existing amide synthesis routes that generate toxic gases or high-molecular-weight by-products.<\/p>\n<p class=\"article-content\">\u201cSome coupling reagents provide higher yield than our method.
