The research led by the Mass General Brigham Cancer Institute team could offer a potential new option for managing the common and challenging complication of chemotherapy-induced thrombocytopenia (CIT) during gastrointestinal cancer treatment. The findings were recently published in the Journal of Clinical Oncology.

Chemotherapy-induced thrombocytopenia (CIT) reduces platelet counts in gastrointestinal cancer patients, forcing dose reductions or treatment delays to prevent severe bleeding. This compromises the effectiveness of chemotherapy and could impact survival outcomes. Despite being a common complication with significant clinical impact, no approved therapies are widely available.

Navigating chemotherapy-induced thrombocytopenia: Insights from the RECITE Phase III trial

Hear from Gerald Soff (Sylvester Comprehensive Cancer Center) about practical strategies for managing CIT in real-world clinical settings.

“The findings from our trial may help improve care for patients who experience persistent CIT, which has historically been a very challenging complication to manage,” explained lead and corresponding author Hanny Al-Samkari (Mass General Brigham Cancer Institute and Harvard Medical School).

The randomized, double-blind trial evaluated avatrombopag versus placebo (1:1) in 47 gastrointestinal cancer patients, from multiple US cancer centers, who presented with persistent CIT (defined as platelets ≤85 × 109/L on day 1 of a chemotherapy cycle despite adequate time to recover from the prior cycle). Participants ranged from 25 to 84 years old; 30% were women, 17% were non-White and 11% were Hispanic or Latino. The primary end point was successful correction of CIT and prevention of recurrence; overall 70% of patients receiving avatrombopag achieved successful treatment of CIT compared with 17% of those receiving placebo. Furthermore, platelet counts were recovered in 83% of patients who received the drug, compared with 46% of those who did not.

An independent monitoring board determined that the trial had met its prespecified stopping criteria for efficacy, resulting in the study ending early. In this trial, no serious adverse events were attributed to avatrombopag and no patients required platelet transfusions.

In terms of next steps, the authors would like further research to be conducted to better understand the long-term impacts of improving CIT management on patient outcomes.

A Phase II trial has demonstrated the safety and effectiveness of the oral drug avatrombopag in managing chemotherapy-induced…

[78
256863,18,135,19,17,256864,7886
post-608054","post","type-post","status-publish","format-standard","has-post-thumbnail","category-health","tag-chemotherapy-induced-thrombocytopenia","tag-eire","tag-health","tag-ie","tag-ireland","tag-randomized-clinical-trials","tag-side-effects
["GET
