{"id":2694,"date":"2025-04-05T03:03:10","date_gmt":"2025-04-05T03:03:10","guid":{"rendered":"https:\/\/www.europesays.com\/uk\/2694\/"},"modified":"2025-04-05T03:03:10","modified_gmt":"2025-04-05T03:03:10","slug":"hormone-free-male-birth-control-pill-enters-clinical-trials-for-the-first-time-sciencealert","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/uk\/2694\/","title":{"rendered":"Hormone-Free Male Birth Control Pill Enters Clinical Trials For The First Time : ScienceAlert"},"content":{"rendered":"<p>A hormone-free male birth control pill is undergoing clinical testing for the first time ever.<\/p>\n<p>The drug, called YCT-529, has performed incredibly well at limiting the production of sperm in mice and non-human primates, all while producing very few side effects.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/new-attempt-at-a-side-effect-free-male-contraceptive-has-proven-effective-in-mice\" target=\"_blank\" rel=\"noopener\">In male mice<\/a>, the unique contraceptive kicks in within a month of use, reducing pregnancies in female mates by close to 100 percent.<\/p>\n<p>Male macaques require a higher dosage of YCT-529, but it also causes a rapid plummet in sperm count without severe side effects.<\/p>\n<p>Importantly, the animals soon regain their fertility when the medicine is stopped. The drug also causes no significant changes in three hormones important for sperm production: testosterone, FSH, or inhibin B.<\/p>\n<p>Similar to female hormonal birth control, male birth control that influences sex hormones can produce unwanted side effects, such as weight gain,  <a href=\"https:\/\/www.sciencealert.com\/depression\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73097\" data-postid=\"156928\" rel=\"nofollow noopener\" target=\"_self\">depression<\/a>, or increased levels of &#8216;bad&#8217; cholesterol. In the past, these unforeseen consequences have <a href=\"https:\/\/www.theatlantic.com\/health\/archive\/2016\/11\/the-different-stakes-of-male-and-female-birth-control\/506120\/\" target=\"_blank\" rel=\"noopener\">stopped some effective forms<\/a> from continuing in human trials.<\/p>\n<p>This has led scientists to try <a href=\"https:\/\/www.sciencealert.com\/on-demand-male-contraceptive-pill-could-switch-your-sperm-off-for-a-day\" target=\"_blank\" rel=\"noopener\">non-hormonal methods<\/a> instead, and so far, they are proving <a href=\"https:\/\/www.nbcnews.com\/health\/mens-health\/male-birth-control-gel-results-promising-rcna153349\" target=\"_blank\" rel=\"noopener\">hopeful<\/a>.<\/p>\n<p>A phase 1  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"156928\" rel=\"nofollow noopener\" target=\"_self\">clinical trial<\/a> for YCT-529 is <a href=\"https:\/\/doi.org\/10.1016\/j.contraception.2024.110809\" target=\"_blank\" rel=\"noopener\">already over and done<\/a>, and while we don&#8217;t yet know the results, they were successful enough that the drug is now entering phase 2, which assesses safety and efficacy. The trial began in <a href=\"https:\/\/www.nzcr.co.nz\/wp-content\/uploads\/2025\/02\/YCT-529-02-NZCR-PISCF-Phase-1b-v5.0_22Nov24_Clean.pdf\" target=\"_blank\" rel=\"noopener\">New Zealand<\/a> in September 2024.<\/p>\n<p>&#8220;A safe and effective male pill will provide more options to couples for birth control,&#8221; <a href=\"https:\/\/twin-cities.umn.edu\/news-events\/first-hormone-free-male-birth-control-pill-clears-another-milestone\" target=\"_blank\" rel=\"noopener\">says<\/a> medicinal chemist Gunda Georg from the University of Minnesota. &#8220;It will allow a more equitable sharing of responsibility for family planning and provide reproductive autonomy for men.&#8221;<\/p>\n<p>Georg&#8217;s lab <a href=\"https:\/\/doi.org\/10.1016\/j.contraception.2024.110809\" target=\"_blank\" rel=\"noopener\">began investigating<\/a> YCT-529 years ago because of its impact on a protein called retinoic acid receptor (RAR) alpha. This receptor interacts with retinoic acid \u2013 a metabolite of  <a href=\"https:\/\/www.sciencealert.com\/what-are-vitamins-and-do-we-really-need-to-take-them\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73116\" data-postid=\"156928\" rel=\"nofollow noopener\" target=\"_self\">vitamin<\/a> A \u2013 to influence cell growth, sperm formation, and embryo development.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/uk\/wp-content\/uploads\/2025\/04\/dcp1107_380.png\" alt=\"Sperm Production\" width=\"380\" height=\"250\" class=\"size-full wp-image-156933\" loading=\"lazy\"\/>Sperm production produced via retinoic acid (RA). (<a href=\"https:\/\/wi.mit.edu\/news\/retinoic-acid-regulates-transitions-mouse-sperm-production\" target=\"_blank\" rel=\"noopener\">MIT<\/a>)<\/p>\n<p>YCT-529 blocks RAR-alpha, and because it only targets this one version of the receptor, it limits possible side effects.<\/p>\n<p>The ongoing research has since received funding from the National Institutes of Health and brings together scientists from Columbia University, the University of Minnesota, and the pharmaceutical company YourChoice Therapeutics.<\/p>\n<p>Years ago, Georg and her colleagues said they would have clinical trials running by 2022. The research is running slower than predicted, but it&#8217;s still heading in a positive direction.<\/p>\n<p>Chief science officer and founder of YourChoice Therapeutics, Nadja Mannowetz, <a href=\"https:\/\/twin-cities.umn.edu\/news-events\/first-hormone-free-male-birth-control-pill-clears-another-milestone\" target=\"_blank\" rel=\"noopener\">says<\/a> their animal studies have &#8220;laid the groundwork for human  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"156928\" rel=\"nofollow noopener\" target=\"_self\">clinical trials<\/a> of YCT-529, which are progressing efficiently.&#8221;<\/p>\n<p>&#8220;With the unintentional pregnancy rate at nearly 50 percent in the US and globally, we need more contraceptive options, particularly for men,&#8221; she <a href=\"https:\/\/twin-cities.umn.edu\/news-events\/first-hormone-free-male-birth-control-pill-clears-another-milestone\" target=\"_blank\" rel=\"noopener\">adds<\/a>.<\/p>\n<p>YCT-529 isn&#8217;t the only non-hormonal male birth control under exploration. A 2024 <a href=\"https:\/\/www.sciencealert.com\/new-type-of-reversible-male-contraception-proves-a-success-in-mouse-study\" target=\"_blank\" rel=\"noopener\">study<\/a>, led by scientists at Baylor College of Medicine, tested another compound, called CDD-2807, which stops male mice from siring litters when it is injected. It is not yet undergoing clinical trials.<\/p>\n<p>The last time a <a href=\"https:\/\/www.sciencealert.com\/expert-explains-why-male-birth-control-options-are-so-limited\" target=\"_blank\" rel=\"noopener\">male contraceptive<\/a> was approved for sale was in the 1980s when scientists figured out how to do a minimally invasive vasectomy.<\/p>\n<p><a href=\"https:\/\/www.contraceptionjournal.org\/article\/S0010-7824(23)00063-X\/abstract\" target=\"_blank\" rel=\"noopener\">A 2023 study<\/a> surveying more than 2,000 male respondents found more than three-quarters were willing to use new contraceptives.<\/p>\n<p>&#8220;Women have shouldered the burden of pregnancy prevention for too long,&#8221; Mannowetz <a href=\"https:\/\/www.clinicaltrialsarena.com\/news\/yourchoice-birth-control-pill\/?cf-view\" target=\"_blank\" rel=\"noopener\">said<\/a> in a recent press release.<\/p>\n<p>&#8220;Data continue to show that men want to help, and they&#8217;re willing to try new birth control options. Data also show women trust them to do so. After nearly two centuries of no innovation in male birth control, it&#8217;s time for change, and we&#8217;re pleased to be a part of it.&#8221;<\/p>\n<p>The study was published in <a href=\"https:\/\/doi.org\/10.1038\/s43856-025-00752-7\" target=\"_blank\" rel=\"noopener\">Nature Communications<\/a>.<\/p>\n","protected":false},"excerpt":{"rendered":"A hormone-free male birth control pill is undergoing clinical testing for the first time ever.
