{"id":427157,"date":"2025-09-15T20:45:10","date_gmt":"2025-09-15T20:45:10","guid":{"rendered":"https:\/\/www.europesays.com\/uk\/427157\/"},"modified":"2025-09-15T20:45:10","modified_gmt":"2025-09-15T20:45:10","slug":"study-ids-better-method-to-detect-genetic-defects-in-sporadic-als","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/uk\/427157\/","title":{"rendered":"Study IDs better method to detect genetic defects in sporadic ALS"},"content":{"rendered":"<p>Long-read DNA sequencing is a more accurate method than short-read sequencing for detecting certain defects in genes associated with an increased risk of sporadic <a href=\"https:\/\/alsnewstoday.com\/what-is-als\/\" target=\"_blank\" rel=\"noopener\">amyotrophic lateral sclerosis<\/a> (ALS), a new study suggests.<\/p>\n<p>A relatively new technique, long-read sequencing can capture data on thousands to hundreds of thousands of nucleotides, the building blocks of DNA. In comparison, the standard short-read sequencing \u2014 while highly accurate up to 300 nucleotides \u2014 can miss certain genetic defects that occur over longer stretches of DNA, the researchers found.<\/p>\n<p>\u201cWe believe that our findings \u2026 will help [elucidate] \u2026 ALS molecular mechanisms and consequently identify potential therapeutic targets,\u201d the researchers wrote. Although more than <a href=\"https:\/\/alsnewstoday.com\/als-and-genetics\/\" target=\"_blank\" rel=\"noopener\">30 genes have been linked<\/a> to the disease, in <a href=\"https:\/\/alsnewstoday.com\/sporadic-als\/\" target=\"_blank\" rel=\"noopener\">sporadic<\/a> cases, about 10% of people \u2014 and not the other 90% or so \u2014 are shown to have mutations in known ALS genes.<\/p>\n<p>Some genetic variations may increase risk of sporadic ALS<\/p>\n<p>Still, there are some genetic variations that may increase susceptibility to sporadic ALS, and knowing which ones may be potentially useful for earlier diagnoses and more effective treatments for these patients.<\/p>\n<p>For example, many genes contain short tandem repeats, or STRs, which are segments of DNA that are repeated multiple times and are essential for regulating gene activity. But sometimes STRs can duplicate or expand and become longer than usual \u2014 a so-called STR expansion \u2014 and disrupt gene function, causing disease.<\/p>\n<p>An STR expansion in the C9ORF72 gene is the most common ALS-causing mutation, and a sizable number of sporadic ALS patients <a href=\"https:\/\/alsnewstoday.com\/news\/some-sporadic-als-patients-have-mutation-causing-other-diseases\/\" target=\"_blank\" rel=\"noopener\">carry STR expansions<\/a>.<\/p>\n<p>The standard approach for detecting STRs is known as short-read sequencing; this method can read up to 300 nucleotides. While the strategy boasts high accuracy and cost-effectiveness, it\u2019s unable to read long sequences.<\/p>\n<p>Long-read sequencing, conversely, is a relatively new technique that excels in capturing lengthier DNA fragments, often spanning thousands to hundreds of thousands of nucleotides. Initially, high error rates and costs limited its use, but recent technological advances have made it more accurate and accessible.<\/p>\n<p>Still, only a few studies have applied long-read sequencing to characterize disease-associated STR expansions. Most studies on ALS-linked STRs were performed using short-read sequencing.<\/p>\n<p>\u201cSTR expansions could represent a valuable starting point to further investigate and clarify the genetic predisposition of sporadic cases,\u201d the researchers wrote.<\/p>\n<p>  Recommended Reading<\/p>\n<p>      <img decoding=\"async\" src=\"https:\/\/www.europesays.com\/uk\/wp-content\/uploads\/2025\/09\/DcotorChart-150x0-c-default.png\" alt=\"A doctor looks at two different sets of brain scans while speaking.\"\/><\/p>\n<p>Researchers conclude long-read sequencing \u2018should be preferred\u2019<\/p>\n<p>Here, the team compared short-read versus long-read sequencing to detect STR expansions in a group of sporadic ALS (sALS) patients.<\/p>\n<p>First, the scientists collected blood samples from 47 adults with sporadic ALS, more than two-thirds of whom (68%) were women. Short-read sequencing was used to detect STR expansions in 12 genes, including\u00a0C9ORF72, associated with various neurodegenerative disorders.<\/p>\n<p>Among the 47 patients, 46 showed STR lengths within the normal range. In the remaining patient, short-read sequencing detected disease-causing STR expansions in the HTT, ATXN2, and\u00a0CACNA1A genes. Intermediate-length STRs were identified in the ATXN3\u00a0and DM1-AS genes of this patient.<\/p>\n<p>When researchers applied long-read sequencing to this patient\u2019s blood sample, however, none of the STR expansions in these genes found by short-read sequencing were detected.<\/p>\n<blockquote class=\"bio-blockquote bio-blockquote--align-left\">\n<p class=\"bio-blockquote-text\">[Short-read sequencing] might fail in identifying pathological [disease-causing] repeats as well as misidentify existing pathological repeats.<\/p>\n<\/blockquote>\n<p>Focusing solely on the C9ORF72 gene, short-read sequencing failed to find any STR expansions among the patient samples.
