{"id":1050240,"date":"2026-09-08T05:11:21","date_gmt":"2026-09-08T05:11:21","guid":{"rendered":"https:\/\/www.europesays.com\/us\/1050240\/"},"modified":"2026-09-08T05:11:21","modified_gmt":"2026-09-08T05:11:21","slug":"scientists-uncover-shared-genetic-roots-of-ocd-tourette-syndrome-and-autism-sciencealert","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/us\/1050240\/","title":{"rendered":"Scientists Uncover Shared Genetic Roots of OCD, Tourette Syndrome, And Autism : ScienceAlert"},"content":{"rendered":"<p>The prenatal to postnatal development of our brains involves a complex cascade of chemicals, branching brain-signaling pathways, and unfolding neural cells \u2013 the evolution of a universe in microcosm.\u00a0<\/p>\n<p>This incredibly intricate development can lead to potentially debilitating conditions like <a href=\"https:\/\/www.sciencealert.com\/ocd-is-a-serious-illness-and-its-more-common-than-you-think\" rel=\"nofollow noopener\" target=\"_blank\">obsessive-compulsive disorder<\/a> (OCD) and chronic tic disorders (CTDs), such as <a href=\"https:\/\/www.sciencealert.com\/tourettes-and-ocd-overlap-and-a-newly-discovered-brain-link-may-explain-why\" rel=\"nofollow noopener\" target=\"_blank\">Tourette disorder<\/a> (TD), which affect up to 2 percent of the population. <\/p>\n<p>Yet relatively little has been known about the underlying biological pathologies of these highly heritable disorders: a measly <a href=\"https:\/\/doi.org\/10.1038\/s41593-026-02419-5\" rel=\"nofollow noopener\" target=\"_blank\">four &#8220;high-confidence&#8221; genes<\/a>.\u00a0<\/p>\n<p>That number has just jumped significantly, as researchers have presented a uniquely detailed genetic analysis of OCD and CTD etiology, identifying 36 genes that substantially increase the risk of developing one or both of these disorders.\u00a0<\/p>\n<p>This research &#8220;dramatically expands the catalog of shared risk genes, reveals biological connections with  <a href=\"https:\/\/www.sciencealert.com\/autism-spectrum-disorder\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73013\" data-postid=\"217847\" rel=\"nofollow noopener\" target=\"_self\">autism<\/a> and  <a href=\"https:\/\/www.sciencealert.com\/schizophrenia\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73022\" data-postid=\"217847\" rel=\"nofollow noopener\" target=\"_self\">schizophrenia<\/a> and highlights the brain circuits that govern impulse control, movement, and habit formation,&#8221; <a href=\"https:\/\/research.rutgers.edu\/news\/researchers-discover-dozens-genes-tied-ocd-and-tic-disorders\" rel=\"nofollow noopener\" target=\"_blank\">explains<\/a> Gary Heiman, a genetic epidemiologist at Rutgers University in New Jersey and one of the study&#8217;s senior co-authors.<\/p>\n<p>Published in the journal <a href=\"https:\/\/doi.org\/10.1038\/s41593-026-02419-5\" rel=\"nofollow noopener\" target=\"_blank\">Nature Neuroscience<\/a>, an immense team of geneticists and neuroscientists, led by researchers at Rutgers University, analyzed newly and previously sequenced exome data \u2013 the DNA associated with making proteins, comprising around <a href=\"https:\/\/www.sciencealert.com\/massive-genetic-study-identifies-a-rare-mutation-that-protects-against-weight-gain\" rel=\"nofollow noopener\" target=\"_blank\">one percent of the genome<\/a> \u2013 of nearly 4,000 individuals who have OCD, CTD, or both. <\/p>\n<p>This includes over 2,400 parent-child trios and more than 1,500 singletons, nearly a doubling of previous sample sizes.\u00a0<\/p>\n<p><img decoding=\"async\" width=\"1085\" height=\"675\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2026\/09\/OCD-1-Large-1-1085x675.jpeg\" alt=\"\" class=\"wp-image-217852\"   loading=\"lazy\"\/>The cohorts included in this research. <a href=\"https:\/\/doi.org\/10.1038\/s41593-026-02419-5\" rel=\"nofollow noopener\" target=\"_blank\">(Wang et al., Nature Neuroscience, 2026)<\/a><\/p>\n<p>These two conditions commonly co-occur: 50 percent of individuals with CTDs also display obsessive-compulsive behaviors, while up to 30 percent of individuals with OCD display a history of tics.<\/p>\n<p>By also using brain maps from humans, rhesus macaques, and mice for comparison, the researchers outlined a neural blueprint for these conditions based spatially on brain regions, the timeline of risk-increasing gene expression, and affected cell types.<\/p>\n<p>They found further evidence to support that these conditions are mediated by dysfunction in the cortico-striato-thalamo-cortical circuit. This expansive brain loop encompasses regions associated with cognition, impulse control, movement, and data processing.\u00a0<\/p>\n<p>Accordingly, the researchers found that these brain regions showed increased expression of risk-associated genes present at prenatal and postnatal stages. These include the cortex, striatum, and thalamus.\u00a0<\/p>\n<p>The <a href=\"https:\/\/www.sciencealert.com\/some-layers-of-your-brain-actually-get-stronger-as-you-age\" rel=\"nofollow noopener\" target=\"_blank\">cortex<\/a> is the outer brain layer associated with core functions like thinking, learning, reasoning, and memory; the <a href=\"https:\/\/www.sciencealert.com\/dopamine-doesnt-work-in-our-brains-quite-the-way-we-thought\" rel=\"nofollow noopener\" target=\"_blank\">striatum<\/a> is a deep brain region that powers decision-making, motivation, and movement; the <a href=\"https:\/\/www.sciencealert.com\/this-part-of-the-brain-could-be-much-more-important-than-we-thought\" rel=\"nofollow noopener\" target=\"_blank\">thalamus<\/a> is the brain&#8217;s relay station, processing various signals.\u00a0<\/p>\n<p>Risk genes are also highly expressed postnatally, in the <a href=\"https:\/\/www.sciencealert.com\/an-unexpected-function-discovered-inside-the-little-brain-at-the-back-of-your-head\" rel=\"nofollow noopener\" target=\"_blank\">cerebellum<\/a>, a coordinator of movements, affecting essential functions like timing and balance.\u00a0<\/p>\n<p><img decoding=\"async\" width=\"1200\" height=\"587\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2026\/09\/OCD-Large-1200x587.jpeg\" alt=\"\" class=\"wp-image-217851\"   loading=\"lazy\"\/>High-risk gene expression in various bodily tissues. <a href=\"https:\/\/doi.org\/10.1038\/s41593-026-02419-5\" rel=\"nofollow noopener\" target=\"_blank\">(Wang et al., Nature Neuroscience, 2026)<\/a><\/p>\n<p>The researchers implicated a type of cell, called telencephalic projecting excitatory neurons, which are vital for brain signaling and communication across brain regions.\u00a0<\/p>\n<p>Furthermore, they detected complex interactions between genes and neural pathways.<\/p>\n<p>&#8220;These genes don&#8217;t act individually,&#8221; <a href=\"https:\/\/research.rutgers.edu\/news\/researchers-discover-dozens-genes-tied-ocd-and-tic-disorders\" rel=\"nofollow noopener\" target=\"_blank\">clarifies<\/a> Jay Tischfield, a geneticist at Rutgers University and one of the study&#8217;s senior co-authors.\u00a0\u00a0<\/p>\n<p>&#8220;They act in networks. And now you can target whole networks, which will make it easier to design new therapies.&#8221;<\/p>\n<p>The magnitude may be substantial. The identified genes carry very large effects; the researchers determined they may increase risk 57-fold on average, with up to a 210-fold increase at the extreme end.\u00a0<\/p>\n<p>What&#8217;s more, this confirms &#8220;substantial genetic overlap between OCD and CTD,&#8221; the researchers <a href=\"https:\/\/doi.org\/10.1038\/s41593-026-02419-5\" rel=\"nofollow noopener\" target=\"_blank\">note<\/a>, as 30 of the 36 identified genes were associated with both OCD and CTD cases. <\/p>\n<p>Finally, the researchers found that some of the risk genes were associated with other neurodevelopmental disorders (NDDs), including <a href=\"https:\/\/www.sciencealert.com\/autism-spectrum-disorder\" rel=\"nofollow noopener\" target=\"_blank\">autism spectrum disorder<\/a>, <a href=\"https:\/\/www.sciencealert.com\/scientists-have-found-a-new-neurodevelopmental-disorder-hidden-in-our-genes\" rel=\"nofollow noopener\" target=\"_blank\">developmental delays<\/a>, and <a href=\"https:\/\/www.sciencealert.com\/schizophrenia\" rel=\"nofollow noopener\" target=\"_blank\">schizophrenia<\/a>, strengthening <a href=\"https:\/\/www.sciencealert.com\/several-psychiatric-disorders-share-the-same-root-cause-study-suggests\" rel=\"nofollow noopener\" target=\"_blank\">previous links between developmental pathologies<\/a>.\u00a0<\/p>\n<p>Importantly, these genes did not show overlap with a control condition, congenital heart disease (CHD), which is also inheritable but independent of neural development.\u00a0<\/p>\n<p>This work also highlights the importance of retroactive data collection and the crucial long-term commitment made by families who offered their DNA samples.\u00a0<\/p>\n<p>&#8220;When we began collecting these samples 20 years ago, we did not yet have the rapid and cost-effective technologies we have today,&#8221; Heiman <a href=\"https:\/\/research.rutgers.edu\/news\/researchers-discover-dozens-genes-tied-ocd-and-tic-disorders\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a>.\u00a0<\/p>\n<p>&#8220;Families volunteered because they wanted to help scientists understand these conditions.
