{"id":253566,"date":"2025-09-25T12:21:13","date_gmt":"2025-09-25T12:21:13","guid":{"rendered":"https:\/\/www.europesays.com\/us\/253566\/"},"modified":"2025-09-25T12:21:13","modified_gmt":"2025-09-25T12:21:13","slug":"breakthrough-gene-therapy-slows-huntingtons-disease-by-75-sciencealert","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/us\/253566\/","title":{"rendered":"Breakthrough Gene Therapy Slows Huntington&#8217;s Disease by 75% : ScienceAlert"},"content":{"rendered":"<p>Researchers in the United Kingdom say they have successfully trialed what could become the world&#8217;s first gene therapy for <a href=\"https:\/\/en.wikipedia.org\/wiki\/Huntington%27s_disease\" rel=\"nofollow noopener\" target=\"_blank\">Huntington&#8217;s disease<\/a> \u2013 a fatal neurodegenerative disorder that is typically inherited.<\/p>\n<p>While the results of the  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"175445\" rel=\"nofollow noopener\" target=\"_self\">clinical trial<\/a> are not yet formally published or peer reviewed, principal investigator and neuroscientist Ed Wild from University College London <a href=\"https:\/\/www.ucl.ac.uk\/news\/2025\/sep\/gene-therapy-appears-slow-huntingtons-disease-progression\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> the gene therapy, called AMT-130, &#8220;changes everything.&#8221;<\/p>\n<p>The highest dose can apparently slow disease progression by as much as 75 percent over three years. It also led to a significant reduction in a biomarker of neurodegeneration, found in cerebrospinal fluid, which usually increases with disease progression.<\/p>\n<p><strong>Related: <a href=\"https:\/\/www.sciencealert.com\/major-breakthrough-experimental-drug-suppresses-huntington-s-protein-first-time-ionis-httrx-gene\" rel=\"nofollow noopener\" target=\"_blank\">Huge Breakthrough as Experimental Drug Is First-Ever to Suppress Huntington&#8217;s Protein<\/a><\/strong><\/p>\n<p>&#8220;On the basis of these results, it seems likely AMT-130 will be the first licensed treatment to slow Huntington&#8217;s disease, which is truly world-changing stuff,&#8221; <a href=\"https:\/\/www.ucl.ac.uk\/news\/2025\/sep\/gene-therapy-appears-slow-huntingtons-disease-progression\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Wild, who works at UCL&#8217;s Huntington&#8217;s Disease Center, the largest Huntington&#8217;s clinical group in Europe.<\/p>\n<p>Huntington&#8217;s is caused by a single defect in the gene HTT, <a href=\"https:\/\/doi.org\/10.1016\/0092-8674(93)90585-E\" rel=\"nofollow noopener\" target=\"_blank\">discovered in 1993<\/a>, which gradually destroys the body&#8217;s ability to function. It does so by making a toxic version of the <a href=\"https:\/\/en.wikipedia.org\/wiki\/Huntingtin\" rel=\"nofollow noopener\" target=\"_blank\">huntingtin protein<\/a>, which attacks neurons in parts of the brain involved in voluntary movement, thinking, and behavior.<\/p>\n<p>When visible symptoms take hold, typically in mid-adulthood, patients generally have just <a href=\"https:\/\/www.mayoclinic.org\/diseases-conditions\/huntingtons-disease\/symptoms-causes\/syc-20356117\" rel=\"nofollow noopener\" target=\"_blank\">10 to 30 years to live<\/a>. A parent with Huntington&#8217;s has a 50 percent chance of passing the disease on to their children.<\/p>\n<p>For roughly a decade, researchers at <a href=\"https:\/\/www.uniqure.com\/pioneers-leaders\" rel=\"nofollow noopener\" target=\"_blank\">uniQure<\/a>, the company behind the <a href=\"https:\/\/doi.org\/10.1002\/emmm.201202291\" rel=\"nofollow noopener\" target=\"_blank\">world&#8217;s first approved gene therapy<\/a>, have been investigating if a similar method could work as a treatment for Huntington&#8217;s.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/09\/AMT130.jpg\" alt=\"Gene Therapy Huntington's\" width=\"638\" height=\"359\" class=\"size-full wp-image-175477\"   loading=\"lazy\"\/>uniQure&#8217;s explanation from 2019 for how it administers AMT-130. (<a href=\"https:\/\/factor-h.org\/2020-9-26-updates-on-clinical-studies-targeting-htt-and-other-topics\" rel=\"nofollow noopener\" target=\"_blank\">uniQure<\/a>)<\/p>\n<p>The company&#8217;s novel drug was tested by neurologists in the United Kingdom, as it involves major brain surgery. The idea is that when high-dose AMT-130 is injected into the brain, neurons take up the custom-made DNA permanently. The genetic information contains instructions that stop cells from making the mutant huntingtin protein.<\/p>\n<p>The researchers expect the single dose to last a lifetime.<\/p>\n<p>&#8220;Trial results come through in numbers and graphs, but behind each data point is an incredible patient who volunteered to undergo major neurosurgery to be treated with the first gene therapy we&#8217;ve ever tested in Huntington&#8217;s disease,&#8221; <a href=\"https:\/\/www.ucl.ac.uk\/news\/2025\/sep\/gene-therapy-appears-slow-huntingtons-disease-progression\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Wild.<\/p>\n<p>&#8220;That is an extraordinary act of bravery for the benefit of humanity.&#8221;<\/p>\n<p>In the phase 1\/2 clinical trial, 29 patients volunteered to be treated with AMT-130, with 17 receiving a high dose and 12 receiving a low dose. Researchers then followed up with a dozen patients from each group over the course of three years.<\/p>\n<p>The high-dose group showed 75 percent less disease progression than participants who did not receive any AMT-130.<\/p>\n<p>That&#8217;s not quite the &#8220;<a href=\"https:\/\/uniqure.gcs-web.com\/news-releases\/news-release-details\/uniqure-announces-positive-topline-results-pivotal-phase-iii\" rel=\"nofollow noopener\" target=\"_blank\">potentially curative results<\/a>&#8221; the company was hoping to deliver, but it could still be a <a href=\"https:\/\/www.sciencealert.com\/gene-therapy-can-restore-hearing-in-adults-first-of-its-kind-trial-shows\" rel=\"nofollow noopener\" target=\"_blank\">life-changing breakthrough<\/a> and the first real treatment for the disease.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/09\/HD_brains.jpg\" alt=\"HD brains\" width=\"642\" height=\"252\" class=\"size-full wp-image-175481\"   loading=\"lazy\"\/>The brain of someone with Huntington&#8217;s Disease (right) next to a health control (left). (<a href=\"https:\/\/sites.uab.edu\/mgraylab\/huntingtons-disease\/\" rel=\"nofollow noopener\" target=\"_blank\">University of Alabama at Birmingham<\/a>)<\/p>\n<p>&#8220;My patients in the trial are stable over time in a way I&#8217;m not used to seeing in Huntington&#8217;s disease,&#8221; <a href=\"https:\/\/www.ucl.ac.uk\/news\/2025\/sep\/gene-therapy-appears-slow-huntingtons-disease-progression\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Wild, &#8220;and one of them is my only medically retired Huntington&#8217;s disease patient who has been able to go back to work.&#8221;<\/p>\n<p>The speed at which AMT-130 has come to clinical trials is impressive. In the span of <a href=\"https:\/\/www.biospace.com\/uniqure-announces-preclinical-proof-of-concept-for-gene-therapy-approach-in-huntington-s-disease-709323?utm_source=chatgpt.com\" rel=\"nofollow noopener\" target=\"_blank\">roughly a decade<\/a>, researchers at uniQure have taken promising preclinical data and replicated the results in animals and then humans.<\/p>\n<p>The company is now conducting further clinical trials in the US and Europe.<\/p>\n<p>The chief medical officer, Walid Abi-Saab, <a href=\"https:\/\/uniqure.gcs-web.com\/news-releases\/news-release-details\/uniqure-announces-positive-topline-results-pivotal-phase-iii\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> uniQure is &#8220;eager to discuss the data with the Food and Drug Administration (FDA)\u2026 later this year, with the goal of submitting a Biologics License Application in the first quarter of 2026.&#8221;<\/p>\n<p>The US FDA has already granted a Breakthrough Therapy designation and a Regenerative Medicine Advanced Therapy designation, both of which could speed up its approval process.
