{"id":30244,"date":"2025-07-01T16:03:12","date_gmt":"2025-07-01T16:03:12","guid":{"rendered":"https:\/\/www.europesays.com\/us\/30244\/"},"modified":"2025-07-01T16:03:12","modified_gmt":"2025-07-01T16:03:12","slug":"ozempic-alternative-ditches-the-needle-and-one-major-side-effect-sciencealert","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/us\/30244\/","title":{"rendered":"Ozempic Alternative Ditches The Needle And One Major Side Effect : ScienceAlert"},"content":{"rendered":"<p>A drug that can be taken orally could soon rival the likes of <a href=\"https:\/\/www.sciencealert.com\/what-is-ozempic-and-why-are-people-saying-its-a-sad-way-to-lose-weight\" target=\"_blank\" rel=\"noopener\">Ozempic<\/a> for treating type 2  <a href=\"https:\/\/www.sciencealert.com\/diabetes\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73018\" data-postid=\"166308\" rel=\"nofollow noopener\" target=\"_self\">diabetes<\/a> and obesity, according to a new study. Although its efficacy hasn&#8217;t yet been tested in humans, the compound could bypass some of the <a href=\"https:\/\/www.sciencealert.com\/ozempic-vs-wegovy-the-risks-and-side-effects-of-semaglutide-drugs\" target=\"_blank\" rel=\"noopener\">harmful side-effects<\/a> associated with similar drugs.<\/p>\n<p>Unlike the famous family of GLP-1 receptor agonist drugs, this new candidate targets the <a href=\"https:\/\/en.wikipedia.org\/wiki\/Beta-2_adrenergic_receptor\" target=\"_blank\" rel=\"noopener\">\u03b22-adrenergic receptor<\/a> (\u03b22AR). Drugs that do this aren&#8217;t new: <a href=\"https:\/\/en.wikipedia.org\/wiki\/Salbutamol\" target=\"_blank\" rel=\"noopener\">salbutamol<\/a> (sold as Ventolin among other brand names) is widely prescribed for use in short stints to treat asthma and chronic obstructive pulmonary disease.<\/p>\n<p><a href=\"https:\/\/en.wikipedia.org\/wiki\/Clenbuterol\" target=\"_blank\" rel=\"noopener\">Clenbuterol<\/a> is also prescribed for asthma in some countries, though it&#8217;s better known as a performance-enhancing drug, <a href=\"https:\/\/doi.org\/10.2165\/11537540-000000000-00000\" target=\"_blank\" rel=\"noopener\">banned by the World Anti-Doping Agency<\/a> for its anabolic effects.<\/p>\n<p>For more than a decade, scientists have been trying to find a way to harness the benefits of \u03b22AR agonists, without setting off their harmful effects. They can stimulate skeletal muscle glucose uptake and burn fat, mimicking the effects of exercise \u2013 but they have been considered too risky for treating diabetes or obesity, because of their tendency to <a href=\"https:\/\/my.clevelandclinic.org\/health\/treatments\/24851-beta-agonist\" target=\"_blank\" rel=\"noopener\">wreak havoc on the heart<\/a>. <\/p>\n<p><strong>Related: <a href=\"https:\/\/www.sciencealert.com\/one-weight-loss-strategy-is-5x-more-effective-than-ozempic-trials-show?utm_source=SA_article&amp;utm_campaign=related_link\" target=\"_blank\" rel=\"noopener\">One Weight Loss Strategy Is 5x More Effective Than Ozempic, Trials Show<\/a><\/strong><\/p>\n<p>The body&#8217;s \u03b22-adrenergic receptors are coupled with G proteins, and when activated for long periods (by \u03b22AR drugs, for instance), the G proteins set off a chain reaction which leads to cardiovascular problems.<\/p>\n<p>&#8220;That would lead to heart rate increases, systolic blood pressure increases, and, eventually, that over-sustained exposure would lead to increases in heart weight and cardiac hypertrophy,&#8221; molecular biologist Shane Wright, from Karolinska Institute in Sweden, <a href=\"https:\/\/www.bioworld.com\/articles\/721572-exercise-mimicking-compound-offers-alternative-to-glp-1-therapies?v=preview\" target=\"_blank\" rel=\"noopener\">told<\/a> Mar de Miguel of Bioworld.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/07\/gr5_lrg-1.jpg\" alt=\"(A) Glucose tolerance test in DIO mice following treatment with clenbuterol, salbutamol, or compounds 15, 21, or 26. (Motso et al., Cell, 2025)<\/p>\n<p>But a campaign led by scientists from the Karolinska Institute, Stockholm University, and the biotech company Atrogi AB has revealed a series of \u03b22AR agonist molecules that can side-step the slippery slope triggered by the G proteins. <\/p>\n<p>By activating only specific pathways, one of these \u2013 dubbed &#8216;compound 15&#8217; \u2013 seems less likely to cause those concerning side effects, though we won&#8217;t know for sure until further clinical trials are carried out.<\/p>\n<p>Unlike GLP-1 receptor agonists like Ozempic, compound 15 doesn&#8217;t suppress appetite or lead to <a href=\"https:\/\/www.sciencealert.com\/experts-are-concerned-drugs-like-ozempic-may-cause-muscle-loss\" target=\"_blank\" rel=\"noopener\">muscle loss<\/a>, which means it may be safer in that regard too. It&#8217;s also much more user-friendly, as a drug that can be swallowed rather than injected.<\/p>\n<p>&#8220;The goal was to try to target this other action that this receptor has expressed in the skeletal muscle, not in the heart, where it is mediating glucose uptake as a very beneficial outlet for lowering blood glucose levels,&#8221; <a href=\"https:\/\/www.bioworld.com\/articles\/721572-exercise-mimicking-compound-offers-alternative-to-glp-1-therapies?v=preview\" target=\"_blank\" rel=\"noopener\">Wright said<\/a>.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/07\/figs4_lrg.jpg\" alt=\"(A\u2013E) (A\u2013C) Changes to body mass, fat mass, and lean mass as well as (D and E) to area under the curves of glucose tolerance and to fasting blood glucose were recorded in diet-induced obesity (DIO) mice for more than 2 months following treatment with different doses of compound 15 (0.03, 0.1, and 0.3 mg\/kg) administered by daily oral gavage. (Motso et al., Cell, 2025)<\/p>\n<p>So far, the newly-designed drug has been tested in lab-grown cells, male mice, rats, beagles, and phase 1  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"166308\" rel=\"nofollow noopener\" target=\"_self\">clinical trials<\/a> (which only test for safety, not efficacy) in healthy and type 2 diabetic men.<\/p>\n<p>&#8220;Lead candidates of the chemical series increased glucose tolerance in preclinical models of diabetes and obesity with little or no increase in contractile force, cardiac lesions, and cardiac hypertrophy,&#8221; the authors <a href=\"https:\/\/doi.org\/10.1016\/j.cell.2025.05.042\" target=\"_blank\" rel=\"noopener\">report<\/a>. They also reversed muscle atrophy induced by GLP-1 drugs.<\/p>\n<p>&#8220;Clinical evaluation demonstrated that our lead candidate was orally bioavailable and safe in both healthy volunteers and type 2 diabetics, underlining its therapeutic potential.&#8221;<\/p>\n<p>Many more tests \u2013 and a catchier name \u2013 will be needed before compound 15 or its relatives are made available as an alternative to GLP-1 drugs.
