First launched in the mid-2000s, LLFS recruited nearly 5,000&#13;<br \/>\nindividuals from 539 families across the U.S. and Denmark \u2014&#13;<br \/>\nfamilies enriched for long life spans \u2014 spanning three&#13;<br \/>\ngenerations, with the oldest members averaging around 90 years at&#13;<br \/>\nrecruitment.<\/p>\n<p>Previously, the project received $68 million over five years in&#13;<br \/>\n2019 from the National Institutes of Health (NIH) National&#13;<br \/>\nInstitute on Aging, with Washington University in St. Louis leading&#13;<br \/>\nas the coordinating and analysis center alongside partner field&#13;<br \/>\nsites at Boston University, Columbia University, the University of&#13;<br \/>\nPittsburgh, and the University of Southern Denmark. Recently, NIH&#13;<br \/>\nextended funding with an additional $80 million over five years,&#13;<br \/>\nallowing LLFS to operate through 2030. To date, more than 5,400&#13;<br \/>\nindividuals have been enrolled and followed through repeated&#13;<br \/>\nin-home assessments over nearly 20 years.<\/p>\n<p>The current phase incorporates cutting-edge&#13;<br \/>\n&#8220;long-read&#8221; whole-genome sequencing with PacBio&#13;<br \/>\ntechnology (up to 7,800 samples), epigenomics, proteomics,&#13;<br \/>\nmetabolomics, and microbiome analysis, enabling detection of rare&#13;<br \/>\nprotective variants, methylation patterns, and other biological&#13;<br \/>\nmarkers that standard approaches often miss.<\/p>\n<p><strong>Impact for Precision Medicine<\/strong><\/p>\n<p>LLFS&#8217;s search for rare, protective genetic variants and the&#13;<br \/>\nintegration of multi-omics with detailed phenotypic and&#13;<br \/>\nlongitudinal data align closely with precision health goals, using&#13;<br \/>\ngenetic signatures to predict disease resistance, tailor preventive&#13;<br \/>\nstrategies, and develop therapies targeted to individual biological&#13;<br \/>\nprofiles. The study&#8217;s cross-generational design also helps&#13;<br \/>\nidentify life-course and environmental factors that may modify&#13;<br \/>\ngenetic risk or resilience.<\/p>\n<p>In an era of overall tightening federal research budgets, this&#13;<br \/>\nsustained and expanded NIH investment underscores the perceived&#13;<br \/>\nhigh impact of LLFS. The findings could directly inform&#13;<br \/>\npersonalized aging interventions, biomarker development, and&#13;<br \/>\ntherapies aimed at extending both lifespan and health span.
