Mounting evidence suggests these tiny molecules could even be put to work protecting the brain from difficult-to-treat conditions like  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"180301\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> and  <a href=\"https:\/\/www.sciencealert.com\/schizophrenia\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73022\" data-postid=\"180301\" rel=\"nofollow noopener\" target=\"_self\">schizophrenia<\/a>.<\/p>\n<p>In a new paper, a team of scientists from the Centre National de la Recherche Scientifique (CNRS) in France explains where the research has gotten us so far, and what lies ahead.<\/p>\n<p>The CNRS experts believe their small size may make them ideal for reaching into and <a href=\"https:\/\/www.sciencealert.com\/new-alzheimers-treatment-clears-plaques-from-brains-of-mice-within-hours\" rel=\"nofollow noopener\" target=\"_blank\">treating the brain<\/a> with fewer side-effects, though so far, the four nanobody therapies available have been approved only for treating other parts of the body.<\/p>\n<p><strong>Related: <a href=\"https:\/\/www.sciencealert.com\/ozempic-literally-came-from-a-monster-and-its-not-alone\" rel=\"nofollow noopener\" target=\"_blank\">Ozempic Literally Came From a Monster \u2013 And It&#8217;s Not Alone<\/a><\/strong><\/p>\n<p><a href=\"https:\/\/en.wikipedia.org\/wiki\/Antibody\" rel=\"nofollow noopener\" target=\"_blank\">Antibodies<\/a> are proteins that our immune system uses to tag unwanted materials in the body \u2013  <a href=\"https:\/\/www.sciencealert.com\/virus\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73053\" data-postid=\"180301\" rel=\"nofollow noopener\" target=\"_self\">viruses<\/a>, toxins, and the like \u2013 as trash to be removed by our biological cleanup crew.<\/p>\n<p><a href=\"https:\/\/en.wikipedia.org\/wiki\/Single-domain_antibody\" rel=\"nofollow noopener\" target=\"_blank\">Nanobodies<\/a> are stripped-down versions of these proteins, and their sleek, stealthy build means they can squeeze between a virus&#8217;s defenses to disarm its most dangerous parts.<\/p>\n<p>Members of the <a href=\"https:\/\/en.wikipedia.org\/wiki\/Camelidae\" rel=\"nofollow noopener\" target=\"_blank\">camelid family<\/a> (including llamas, camels, and alpacas) naturally produce smaller antibodies than humans. Scientists have further refined these in the lab to be about 10 times smaller than the common Y-shaped Immunoglobulin G antibody.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/11\/gr1_lrg-4-642x336.jpg\" alt=\"A Unique Characteristic in Camels And Llamas Could Be Used to Treat Alzheimer's\" width=\"642\" height=\"336\" class=\"size-medium wp-image-180380\"   loading=\"lazy\"\/>Nanobodies have unique features for targeting the central nervous system. (Lafon et al., Trends in Pharm. Sci., 2025)<\/p>\n<p>While sharks are also known to produce nanobodies, it&#8217;s our mammalian relatives whose biological machinery is best positioned to supplement our comparatively clunky immune systems.<\/p>\n<p>Nanobodies made from camelids have already shown promise in protecting humans from <a href=\"https:\/\/www.sciencealert.com\/nano-scale-multidomain-antibodies-from-camelids-used-for-universal-influenza-vaccine\" rel=\"nofollow noopener\" target=\"_blank\">influenza A and B<\/a>, <a href=\"https:\/\/www.sciencealert.com\/llamas-hide-a-powerful-weapon-that-can-fight-deadly-virus-strains\" rel=\"nofollow noopener\" target=\"_blank\">the gastro-causing norovirus<\/a>, <a href=\"https:\/\/www.sciencealert.com\/llama-blood-could-play-a-role-in-helping-people-fight-off-coronavirus-infections\" rel=\"nofollow noopener\" target=\"_blank\">COVID-19<\/a>, and <a href=\"https:\/\/www.sciencealert.com\/a-hidden-defense-inside-llamas-could-be-the-secret-to-fighting-hiv\" rel=\"nofollow noopener\" target=\"_blank\">even HIV<\/a>.<\/p>\n<p>Until recently, it seemed nanobodies could not be used to treat brain disorders because human kidneys eliminate them from the bloodstream before they even get a chance to reach their target destination. It also seemed they may not readily cross the <a href=\"https:\/\/www.sciencealert.com\/scientists-think-theyre-on-the-verge-of-breaching-the-blood-brain-barrier\" rel=\"nofollow noopener\" target=\"_blank\">blood brain barrier<\/a>: a decisive checkpoint for any drugs targeting the brain.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/spark-into-space-comp\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/10\/Mid-Article-Promo-Astro-642x272.jpg\" alt=\"Win a $10,000 Space Coast Adventure Holiday\" width=\"642\" height=\"272\" class=\"alignnone wp-image-177074 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>But newer studies have overcome these challenges, with animal model lab experiments showing that engineered nanobodies can cross the blood brain barrier to target and eliminate key markers of Alzheimer&#8217;s disease thought to drive symptoms, <a href=\"https:\/\/doi.org\/10.1038\/s41467-025-58383-4\" rel=\"nofollow noopener\" target=\"_blank\">tau<\/a> and <a href=\"https:\/\/doi.org\/10.3390\/ijms24032632\" rel=\"nofollow noopener\" target=\"_blank\">amyloid beta<\/a>.<\/p>\n<p>&#8220;Camelid nanobodies open a new era of biologic therapies for brain disorders and revolutionize our thinking about therapeutics,&#8221; <a href=\"https:\/\/www.scimex.org\/newsfeed\/camels-and-llamas-could-hold-the-key-to-treating-future-brain-disorders\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Philippe Rondard, neuropharmacologist at CNRS. &#8220;We believe they can form a new class of drugs between conventional antibodies and small molecules.&#8221;<\/p>\n<p>But before these drugs get anywhere near a living human brain, scientists will need to evaluate their stability, proper folding, and ensure the absence of aggregation, Rondard says.<\/p>\n<p>&#8220;These are highly soluble small proteins that can enter the brain passively,&#8221; <a href=\"https:\/\/www.scimex.org\/newsfeed\/camels-and-llamas-could-hold-the-key-to-treating-future-brain-disorders\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> functional genomicist Pierre-Andr\u00e9 Lafon. &#8220;By contrast, small-molecule drugs that are designed to cross the blood-brain barrier are hydrophobic in nature, which limits their bioavailability, increases the risk of off-target binding, and is linked to side effects.&#8221;<\/p>\n<p>We also need to know more about exactly how they&#8217;re crossing the blood brain barrier, and how long they hang around in the brain, to ensure proper dosage.
