{"id":381903,"date":"2025-11-16T01:08:18","date_gmt":"2025-11-16T01:08:18","guid":{"rendered":"https:\/\/www.europesays.com\/us\/381903\/"},"modified":"2025-11-16T01:08:18","modified_gmt":"2025-11-16T01:08:18","slug":"one-of-our-biggest-hopes-for-alzheimers-treatment-doesnt-seem-to-work-sciencealert","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/us\/381903\/","title":{"rendered":"One of Our Biggest Hopes For Alzheimer&#8217;s Treatment Doesn&#8217;t Seem to Work : ScienceAlert"},"content":{"rendered":"<p>For decades, scientists have targeted the sticky protein clumps that amass in <a href=\"https:\/\/www.sciencealert.com\/alzheimer-s-disease\" rel=\"nofollow noopener\" target=\"_blank\">Alzheimer&#8217;s brains<\/a> as a potential way of treating the disease, but they may have been off the mark.<\/p>\n<p>A new study shows that clearing away the amyloid-beta clumps doesn&#8217;t appear to repair key brain functions.<\/p>\n<p>In particular, it doesn&#8217;t restore the brain&#8217;s mechanisms for clearing out waste, known as the glymphatic system. This system is <a href=\"https:\/\/doi.org\/10.1007\/s11604-017-0617-z\" rel=\"nofollow noopener\" target=\"_blank\">known to be impaired<\/a> in people with  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"181214\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a>, and would usually help with getting rid of excess amyloid-beta plaques, via <a href=\"https:\/\/www.sciencealert.com\/scientists-discover-hidden-pulse-in-the-brain-that-may-warn-of-dementia\" rel=\"nofollow noopener\" target=\"_blank\">waves of cerebrospinal fluid<\/a> (CSF).<\/p>\n<p>The study comes from researchers at the Osaka Metropolitan University in Japan, who tested the new Alzheimer&#8217;s drug <a href=\"https:\/\/www.sciencealert.com\/new-alzheimers-treatment-slows-decline-but-comes-at-a-high-cost\" rel=\"nofollow noopener\" target=\"_blank\">lecanemab<\/a> on 13 people with the disease. <a href=\"https:\/\/en.wikipedia.org\/wiki\/Magnetic_resonance_imaging\" rel=\"nofollow noopener\" target=\"_blank\">Magnetic resonance imaging<\/a> ( <a href=\"https:\/\/www.sciencealert.com\/magnetic-resonance-imaging\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73063\" data-postid=\"181214\" rel=\"nofollow noopener\" target=\"_self\">MRI<\/a>) scans were used to look at the effects on the brain.<\/p>\n<p><strong>Related: <a href=\"https:\/\/www.sciencealert.com\/your-heart-health-in-middle-age-could-predict-your-later-dementia-risk\" rel=\"nofollow noopener\" target=\"_blank\">Your Heart Health in Middle Age Could Predict Your Later Dementia Risk<\/a><\/strong><\/p>\n<p>&#8220;Even when amyloid-beta is reduced by lecanemab, impairment of the glymphatic system may not recover within the short-term,&#8221; <a href=\"https:\/\/www.omu.ac.jp\/en\/info\/research-news\/entry-95626.html\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> medical researcher Tatsushi Oura, of Osaka Metropolitan University.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/11\/GlymphaticSystem.jpg\" alt=\"Glymphatic system scan\" width=\"642\" height=\"646\" class=\"wp-image-181217 size-full\"   loading=\"lazy\"\/>A brain scan showing the diffusion of water, which was used to assess how well the glymphatic system was working. (Osaka Metropolitan University)<\/p>\n<p>The study highlights yet again the multifaceted nature of Alzheimer&#8217;s, which is associated with a wide range of potential causes, <a href=\"https:\/\/www.sciencealert.com\/these-15-factors-are-linked-to-early-dementia-risk-a-major-study-found\" rel=\"nofollow noopener\" target=\"_blank\">risk factors<\/a>, and symptoms. It&#8217;s not clear exactly how the disease starts, but various triggers are likely contributing.<\/p>\n<p>When it comes to the buildup of amyloid-beta and another brain protein <a href=\"https:\/\/www.sciencealert.com\/sky-high-levels-of-alzheimers-protein-found-in-newborns\" rel=\"nofollow noopener\" target=\"_blank\">called tau<\/a>, these are tell-tale signs of the disease \u2013 and yet scientists are still trying to figure out whether they&#8217;re causing Alzheimer&#8217;s, caused by Alzheimer&#8217;s, or both.<\/p>\n<p>In this study, lecanemab worked to the extent that it reduced amyloid-beta levels in the brain, but measures of glymphatic system function did not improve after three months. In other words, the drug doesn&#8217;t seem to reverse the damage <a href=\"https:\/\/www.sciencealert.com\/alzheimers-may-hijack-your-cells-clocks-hinting-at-new-treatments\" rel=\"nofollow noopener\" target=\"_blank\">done by Alzheimer&#8217;s<\/a>, at least in terms of the brain&#8217;s waste recycling.<\/p>\n<p>&#8220;Disease-modifying therapy can reduce plaque burden and slow further cognitive worsening but does not restore lost function, likely reflecting the fact that neuronal damage and clearance system deficits have already been well established,&#8221; the researchers <a href=\"https:\/\/doi.org\/10.1002\/jmri.70118\" rel=\"nofollow noopener\" target=\"_blank\">write<\/a> in their published paper.<\/p>\n<p>The study adds more nuance to the increasingly complex picture of Alzheimer&#8217;s, and how it <a href=\"https:\/\/www.sciencealert.com\/alzheimers-and-cancer-may-soon-be-treated-with-sounds-we-cant-hear\" rel=\"nofollow noopener\" target=\"_blank\">might be tackled<\/a>. It&#8217;s not the first study to find that clearing plaques doesn&#8217;t substantially improve Alzheimer&#8217;s, so some researchers are now suspecting the plaques are a <a href=\"https:\/\/www.sciencealert.com\/there-s-even-more-reason-now-to-stop-blaming-amyloid-plaques-for-alzheimer-s\" rel=\"nofollow noopener\" target=\"_blank\">consequence of the disease<\/a>, <a href=\"https:\/\/www.sciencealert.com\/we-might-been-wrong-what-kills-brain-cells-alzheimer-s-disease-beta-amyloid-tau-protein-app\" rel=\"nofollow noopener\" target=\"_blank\">not the cause<\/a>.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/new-alzheimers-treatment-slows-decline-but-comes-at-a-high-cost\" rel=\"nofollow noopener\" target=\"_blank\">Past trials<\/a> have shown that lecanemab does work at slowing down the steady march of Alzheimer&#8217;s disease, but the drug functions best when administered at an early stage.
