{"id":626238,"date":"2026-03-02T06:15:17","date_gmt":"2026-03-02T06:15:17","guid":{"rendered":"https:\/\/www.europesays.com\/us\/626238\/"},"modified":"2026-03-02T06:15:17","modified_gmt":"2026-03-02T06:15:17","slug":"semaglutide-may-reverse-damage-caused-by-osteoarthritis-study-suggests-sciencealert","status":"publish","type":"post","link":"https:\/\/www.europesays.com\/us\/626238\/","title":{"rendered":"Semaglutide May Reverse Damage Caused by Osteoarthritis, Study Suggests : ScienceAlert"},"content":{"rendered":"<p>Medical researchers have discovered that semaglutide may reverse the debilitating tissue damage caused by osteoarthritis, the world&#8217;s <a href=\"https:\/\/www.sciencealert.com\/radiation-therapy-reduces-pain-from-the-most-common-form-of-arthritis\" rel=\"nofollow noopener\" target=\"_blank\">most common form of arthritis<\/a>.<\/p>\n<p>The drug behind Ozempic and Wegovy is best known for treating type 2  <a href=\"https:\/\/www.sciencealert.com\/diabetes\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73018\" data-postid=\"193311\" rel=\"nofollow noopener\" target=\"_self\">diabetes<\/a> and helping people lose weight. But the new study found semaglutide appears to protect joints in mice through a mechanism that&#8217;s not about easing pressure through weight loss.<\/p>\n<p>Instead, the drug reprograms the metabolism of cells that synthesize and maintain healthy cartilage, allowing them to generate more energy.<\/p>\n<p>&#8220;This work not only highlights the potential off-target effect of semaglutide as an effective drug to treat metabolic osteoarthritis,&#8221; the team from China and the US <a href=\"https:\/\/doi.org\/10.1016\/j.cmet.2026.01.008\" rel=\"nofollow noopener\" target=\"_blank\">writes in their published paper<\/a>, &#8220;but also reveals a weight loss-independent repair mechanism.&#8221;<\/p>\n<p>In addition to mouse experiments, the researchers also conducted a randomized human trial to assess the effects of semaglutide on pain, mobility, and tissue degradation in individuals with osteoarthritis and obesity.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2026\/03\/Osteoarthritis-642x482.png\" alt=\"\" width=\"642\" height=\"482\" class=\"size-medium wp-image-193313\"   loading=\"lazy\"\/>Illustrated comparison of a normal knee and a knee with osteoarthritis. These conditions can affect just about every tissue in the body, including bone, cartilage, and <a href=\"https:\/\/my.clevelandclinic.org\/health\/body\/25002-synovial-membrane\" rel=\"nofollow noopener\" target=\"_blank\">synovium<\/a> (the lubricating membrane lining the inner surfaces of joints).<\/p>\n<p>Semaglutide-based drugs like <a href=\"https:\/\/www.sciencealert.com\/what-is-ozempic-and-why-are-people-saying-its-a-sad-way-to-lose-weight\" rel=\"nofollow noopener\" target=\"_blank\">Ozempic and its kin<\/a> work by mimicking glucagon-like peptide 1 (GLP-1), a natural hormone that stimulates insulin release to manage blood sugar levels. This hormone also tells the brain that mealtime has concluded, facilitating an appetite-suppressing effect.<\/p>\n<p>Therefore, it seems obvious that weight loss eases osteoarthritis by reducing load-bearing forces on the joints. But this isn&#8217;t always the case. Existing osteoarthritis therapies are generally palliative. So while they may alleviate the symptoms, they don&#8217;t address the underlying cause.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/newsletter?utm_source=promo_generic_health\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/11\/Generic-Health-Promo-Final-642x273.jpg\" alt=\"Subscribe to ScienceAlert's free fact-checked newsletter\" width=\"642\" height=\"273\" class=\"alignnone wp-image-182810 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>But semaglutide appears to affect joint health at a fundamental level. In mice and humans with <a href=\"https:\/\/www.sciencealert.com\/ozempic-gives-strong-relief-to-severe-knee-pain-clinical-trial-suggests\" rel=\"nofollow noopener\" target=\"_blank\">obesity and osteoarthritis<\/a>, treatment with semaglutide reduced pain and decreased cartilage degeneration. Mice also had fewer bone spurs and less severe lesions in their joint membranes.<\/p>\n<p>When the researchers compared cartilage from treated and untreated mice, they detected changes in the expression of nearly 8,300 proteins.<\/p>\n<p>They included a crucial control: a &#8220;pair-feeding&#8221; group that ate the same amount as the semaglutide-treated mice. Even with comparable weight changes, the pair-feeding group did not receive the same cartilage protection, suggesting a weight-loss-independent effect on the joint itself.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2026\/03\/0910_Oateoarthritis_Hip_A-642x566.png\" alt=\"\" width=\"642\" height=\"566\" class=\"size-medium wp-image-193314\"   loading=\"lazy\"\/>Osteoarthritis causes damage to multiple body tissues, leading to pain and loss of mobility. (<a href=\"https:\/\/commons.wikimedia.org\/wiki\/File:0910_Oateoarthritis_Hip_A.png#mw-jump-to-license\" rel=\"nofollow noopener\" target=\"_blank\">Wikimedia Commons\/CFCF\/CC BY-SA 4.0<\/a>)<\/p>\n<p>Yet it seems the most important biological pathway altered by semaglutide is the &#8220;GLP-1R-AMPK-PFKFB3 axis,&#8221; a cascade of compounds that affects how various cells produce energy.<\/p>\n<p>By initiating this cascade of enzymes, semaglutide alters the metabolic processes used by <a href=\"https:\/\/en.wikipedia.org\/wiki\/Chondrocyte\" rel=\"nofollow noopener\" target=\"_blank\">chondrocytes<\/a> (cells found in healthy cartilage), making them more efficient and allowing them to survive.<\/p>\n<p>As the researchers revealed, a process called glycolysis generated most of the energy for <a href=\"https:\/\/www.sciencealert.com\/new-breakthrough-to-restore-aging-joints-could-help-treat-osteoarthritis\" rel=\"nofollow noopener\" target=\"_blank\">chondrocytes<\/a> in osteoarthritis-affected mice.<\/p>\n<p>Glycolysis doesn&#8217;t require oxygen, so it supplies energy quickly during stressful, high-intensity exercise, like the morning sprint to catch the garbage truck. Yet, due to the lack of oxygen, glycolysis produces only two net ATP (energy-providing) molecules per glucose molecule.<\/p>\n<p>But after semaglutide treatments, a process called oxidative phosphorylation (OXPHOS) became the preferred metabolic pathway for chondrocytes. Unlike glycolysis, OXPHOS uses oxygen and produces much more ATP, up to 36 molecules per glucose molecule.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2026\/03\/Hypertrophic_Zone_of_Epiphyseal_Plate.jpg\" alt=\"\" width=\"642\" height=\"903\" class=\"wp-image-193315\"\/>Chondrocytes are cells responsible for synthesizing and maintaining collagen. Hunt<\/a>\/Public Domain)<\/p>\n<p>To see if any of this might translate to humans, the researchers recruited 20 individuals, aged 50-75, with both obesity and osteoarthritis, and randomly divided them into two groups. One group received sodium hyaluronate (HA), a joint-lubricating, especially effective form of <a href=\"https:\/\/www.sciencealert.com\/new-biomaterial-could-finally-repair-damaged-cartilage-in-injured-joints\" rel=\"nofollow noopener\" target=\"_blank\">hyaluronic acid<\/a>. The other group received both HA and semaglutide.<\/p>\n<p>At the end of the 24-week treatment period, the HA+semaglutide group experienced lower osteoarthritis pain scores and substantial improvements in knee function.  <a href=\"https:\/\/www.sciencealert.com\/magnetic-resonance-imaging\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73063\" data-postid=\"193311\" rel=\"nofollow noopener\" target=\"_self\">MRI<\/a> analysis also revealed thicker cartilage and recent cartilage growth in the inner joint areas that bear weight and absorb shock from daily movements.<\/p>\n<p>The findings from this study may impact global healthcare outcomes. Osteoarthritis affects around <a href=\"https:\/\/www.sciencealert.com\/the-best-medicine-for-joint-pain-isnt-what-you-think-says-expert\" rel=\"nofollow noopener\" target=\"_blank\">600 million people globally<\/a> and is forecast to afflict one billion people by 2050. It&#8217;s also increasing among <a href=\"https:\/\/www.sciencealert.com\/osteoarthritis-is-appearing-in-younger-adults-triggering-decades-of-discomfort\" rel=\"nofollow noopener\" target=\"_blank\">younger, active populations<\/a>, leading to decades of pain and disability.<\/p>\n<p>The work adds to growing evidence that GLP-1 drugs may have <a href=\"https:\/\/www.sciencealert.com\/not-just-weight-loss-8-conditions-drugs-like-wegovy-might-treat\" rel=\"nofollow noopener\" target=\"_blank\">benefits beyond weight loss<\/a>, and it sharpens the search for new osteoarthritis treatments that target metabolism inside the joint.<\/p>\n<p><strong>Related: <a href=\"https:\/\/www.sciencealert.com\/will-exercise-help-your-osteoarthritis-a-new-review-has-surprising-findings\" rel=\"nofollow noopener\" target=\"_blank\">Will Exercise Help Your Osteoarthritis?
