Now researchers have come up with a rather clever alternative: dental floss.<\/p>\n<p>Led by a team from Texas Tech University, the researchers first identified an often overlooked surface in the mouth as an entry point for vaccines: the <a href=\"https:\/\/en.wikipedia.org\/wiki\/Junctional_epithelium\" target=\"_blank\" rel=\"noopener\">junctional epithelium<\/a> (JE), which sits where the gums meet the teeth.<\/p>\n<p>The JE is leaky by design, because it allows <a href=\"https:\/\/www.sciencealert.com\/this-centuries-old-hack-is-a-proven-way-to-boost-your-health-and-immunity\" target=\"_blank\" rel=\"noopener\">immune cells<\/a> to move around and defend the oral cavity from bacterial attack. The researchers wondered whether this could also make this part of the mouth a suitable target for vaccines.<\/p>\n<p>&#8220;We hypothesized that this leakiness of the JE could expedite the entry of vaccine antigens, and the abundance of immune cells in this microenvironment could elicit an adaptive immune response,&#8221; the researchers <a href=\"https:\/\/dx.doi.org\/10.1038\/s41551-025-01451-3\" style=\"letter-spacing: -0.45px;\" target=\"_blank\" rel=\"noopener\">write<\/a> in their recently published paper.<\/p>\n<p><strong>Related: <a href=\"https:\/\/www.sciencealert.com\/which-arm-gets-vaccinated-could-play-a-role-in-your-immune-response?utm_source=SA_article&amp;utm_campaign=related_link\" target=\"_blank\" rel=\"noopener\">Which Arm Gets Vaccinated Could Play a Role in Your Immune Response<\/a><\/strong><\/p>\n<p>Delivering medications to the JE is complicated by the tissue&#8217;s seclusion within the gumline, which is why it hasn&#8217;t really been considered as a potential location <a href=\"https:\/\/doi.org\/10.1016\/j.jconrel.2014.05.060\" target=\"_blank\" rel=\"noopener\">for introducing vaccines<\/a>.<\/p>\n<p>That&#8217;s where floss comes in, which is perfect for getting into tight spaces.<\/p>\n<p>The researchers tested their hypothesis by coating dental floss with different <a href=\"https:\/\/www.sciencealert.com\/the-pfizer-and-moderna-vaccines-are-a-breakthrough-here-s-how-they-work\" target=\"_blank\" rel=\"noopener\">types of vaccines<\/a> and testing them on mice across a period of several weeks.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.europesays.com\/us\/wp-content\/uploads\/2025\/07\/MouseFloss.jpg\" alt=\"Mouse flossing diagram\" width=\"642\" height=\"263\" class=\"wp-image-169026 size-full\"   loading=\"lazy\"\/>The researchers targeted a specific part of the mouth in mice. (Ingrole et al., Nat. Biomed. Eng., 2025)<\/p>\n<p>There were numerous encouraging indicators: strong <a href=\"https:\/\/www.sciencealert.com\/which-arm-gets-vaccinated-could-play-a-role-in-your-immune-response\" target=\"_blank\" rel=\"noopener\">immune responses<\/a> were observed in the mouth and throughout the bodies of the mice, and the floss vaccine was effective in protecting mice against a later  <a href=\"https:\/\/www.sciencealert.com\/what-is-influenza\" class=\"lar_link lar_link_outgoing\" data-linkid=\"103478\" data-postid=\"169023\" rel=\"nofollow noopener\" target=\"_self\">flu<\/a> infection.<\/p>\n<p>Further tests were conducted on human volunteers using dye rather than a vaccine, since  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"169023\" rel=\"nofollow noopener\" target=\"_self\">clinical trials<\/a> are still some way off. The amount of dye that reached the JE target via flossing was enough to suggest that this really could work in us too.<\/p>\n<p>&#8220;These findings establish floss-based vaccination as a simple, needle-free strategy that enhances vaccine delivery and immune activation compared with existing mucosal immunization methods,&#8221; <a href=\"https:\/\/dx.doi.org\/10.1038\/s41551-025-01451-3\" target=\"_blank\" rel=\"noopener\">write<\/a> the researchers.<\/p>\n<p>It&#8217;s not the first time scientists have looked beyond the thorough cleaning job that floss does. Earlier this year <a href=\"https:\/\/www.sciencealert.com\/new-smart-dental-floss-can-detect-your-stress-from-saliva\" target=\"_blank\" rel=\"noopener\">research showed<\/a> that floss could also be adapted to measure stress levels in the body, potentially giving doctors another useful diagnostic tool.<\/p>\n<p>As well as being potentially less scary for those with a phobia of needles, floss-based vaccines would be easier to transport and store. Doctors and nurses wouldn&#8217;t be needed to administer them, and they could even be delivered in the post \u2013 a real advantage in the case of pandemics <a href=\"https:\/\/www.sciencealert.com\/here-s-why-some-covid-19-strains-are-spreading-faster-than-others\" target=\"_blank\" rel=\"noopener\">such as COVID-19<\/a>.<\/p>\n<p>Researchers have previously tried to <a href=\"https:\/\/doi.org\/10.1016\/j.jconrel.2021.03.017\" target=\"_blank\" rel=\"noopener\">deliver vaccines orally<\/a> under the tongue and in the cheek, but not enough of the vaccine penetrates the tissue into circulation. While there&#8217;s still a lot of work to do with this new approach, the potential is clear.<\/p>\n<p>&#8220;Due to drawbacks associated with injectable vaccines, such as pain and needle phobia, potential transmission of blood-borne pathogens from unsafe injection practices and the limited activation of mucosal immunity, there is a need for alternative approaches for vaccine delivery,&#8221; <a href=\"https:\/\/dx.doi.org\/10.1038\/s41551-025-01451-3\" target=\"_blank\" rel=\"noopener\">write<\/a> the researchers.<\/p>\n<p>The research has been published in <a href=\"https:\/\/dx.doi.org\/10.1038\/s41551-025-01451-3\" target=\"_blank\" rel=\"noopener\">Nature Biomedical Engineering<\/a>.<\/p>\n","protected":false},"excerpt":{"rendered":"A fear of needles is a common reason for avoiding vaccines, even among many adults.
